Introduction
Caenorhabditis elegans is a very important study model that is used for many genetic study because of its features like transparent, easy to handle, short life cycle etc. The feeding behavior is very important in the feeding method of the RNAi silencing experiment that use C. elegans to study genetic. The feeding of bacteria could influence the efficiency of the RNAi silencing. The feeding behavior of C. elegans involves uptake of bacteria, pharyngeal pumping, and isthmus peristalsis.To soak the bacteria inside the C. elegans require pharyngeal pumping to initiate the muscle contraction so that the bacteria can be ingested inside, then followed by the contraction of isthmus …show more content…
Equipment such as inverted DIC microscope, a standard dissecting microscope, needle holder etc. are also required. The constructed plasmid that has ccb-1 inserted and a empty vector for control are both injected into the gonad of two group of the young adult hermaphrodites using microinjection needle (see Figure. 6 and 7) using microinjection needles under the inverted DIC microscope (Eisenmann and Wnt, 2005; Strange, 2006). Injection into the gonad of the young adult hermaphrodites because more affected progeny will be produced if inject into gonad (Strange, 2006). And put the injected worm into a new plate every 24 hours to observe the progeny at different time (Eisenmann and Wnt, 2005). Then using a fluorescence dissection microscope to see the GFP that made from mCherry to observe the result from injection whether the gene has been successfully injected into the worm
