Comparison between MPN and E coli Petri film:
The use of E.coli petrifilm seems more reliable than MPN since the value of MPN is less than E.coli petrifilm in the same group. It showed that MPN method was underestimated the total coliform and E.coli numbers.
The Most Probable Number test methods described in this Practical provide guidelines for detection and enumeration of bacteria of food and water origin. Then we can prevent food borne infections and diseases occurring more commonly.
Disadvantages of MPN Techniques:
MPN procedure takes very long time for the confirmed test result. In MPN the results are probability calculations and cannot be accurate.MPN requires more glass wares and media. False positive results are of common occurrence.
Advantages of MPN Techniques:
Interpretation of the results requires minimal experience and training as results can be got by simply observing for the presence of gas or no gas. Also, water samples with high turbidity can be analyzed, since there is no apparent deleterious effect. Furthermore, because of the dilutions used in the range of 1:0 or 1:100, toxic substances present in the sample can be diluted out. Finally, MPN technique is the effective method for analyzing samples such as muds, sludges, sediments etc. Disadvantages of Petri film include:
There is no visual difference between the different kinds of bacteria growing on the Petri film and you can't do any testing on the colonies. No classifying as bacilli or cocci, no gram-staining, no isolating a single type of bacteria.

The advantages of Petri film include
Ready to use as it does not need preparation required. Also long shelf life when shielded from moisture and humidity .Furthermore, simple to use and interpret. Then, it is Built-in biochemical confirmation. Finally, it is smaller and less likely to be damaged than agar

Another factor that is resposinble for my imporvement is the class disscusions. Mrs Philips freuqnetly asaked questons in class about the readings in order to prompt class discussions. The questions raised by mrs philips along with the class discussions that occred due to her questioning allowed for my critical thinking to devlop because both gave me the oppurtunity to think and reflect on what it was that I read. The second area that I feel I have imporved the most in is writing. In the course we did a lot of written work which gave me the oppurtunity to work on my writing. The other reason why my writing was able to imporbe is due to all the feedback I recieved on my work. Hence, if the results of the experiment reject my null hypothesis, it means the the size of the horn on male will affect its mating rate, the larger horn will attract females more and get more mates, also the larger horn cause higher pressure to rival males. If the results doesn’t change, which means my null hypothesis fails to be rejected, the horn doesn’t affect the mate between male and female beetles. Brandon Bryn (2012) tested that in male Rhinoceros beetle, horn size signals healthy mate. This observation rejected my null hypothesis, I will further investigate the detail in experiment...

Clostridium Botulinum Journal Review

...JOURNAL REVIEW FOOD SAFETY & SANITATION INFANT BOTULISM IN HONEY By: Angela Indryana (03420100086) Zefanya Tjokrodiredjo (03420100087) FOOD TECHNOLOGY DEPARTMENT FACULTY OF INDUSTRIAL TECHNOLOGY UNIVERSITAS PELITA HARAPAN KARAWACI 2012 CHAPTER I INTRODUCTION Honey is a natural food, mainly composed of a complex mixture of carbohydrates and other minor substances, such as organic acids, amino acids, proteins, minerals, vitamins, and lipids. It has two sources of contamination with microorganisms: primary sources include pollen, the digestive tracts of honey bees, dust, air, soil and nectar; secondary sources are those arising from honey manipulation by people, they include air, food handlers, cross-contamination, equipment and buildings. Spores of Clostridium botulinum may be found in honey, usually at low levels. C. botulinum spores are found in soil and dust and hence, can contaminate different agricultural products. Honey is a recognized vehicle for C. botulinum spores. The presence of spores of Clostridium is especially dangerous for infants and small children. Although the high sugar content, low pH, low protein concentration, and the existence of different oxidases and other antimicrobial substances in honey may cause the spores unable to germinate and grow in the product, the microenvironment may remain anaerobic as a consequence of high viscosity. Yet existing spores may not be removed from honey or destroyed without spoiling the product, thus...

Distillation

...ABSTARCT Determination of boiling point and melting point of the organic liquids, in practice determining the boiling point of ethyl and Isopropyl Alcohol and the melting point of benzoic acid and Urea. However, the boiling point of a liquid is the temperature at which the pressure of the vapor above a liquid equals the existing pressure. As we heat a liquid, the pressure of the vapor above the liquid slowly increases. When this pressure equals the pressure presented in the container, the liquid begins to boil– the liquid turns to vapor. Many factors affect the boiling point and melting point of an organic compound, and one of which is the molecular structure. Purity and branching also affects the boiling and melting point of a compound. There are three trends that affect the boiling and melting points and one of these trends is the relative strength of the four intermolecular attractions is factor significally affects the boiling point and the melting point of a compound. INTRODUCTION Internationally, boiling points is a useful physical property for demonstrating the purity of an organic compound. Boiling point is the temperature at which the vapor pressure of a liquid equal atmospheric pressure or some other applied pressure. A boiling point is commonly measured when the liquid heated. The boiling temperature is measured as distillation vapor covers the bulb of a thermometer suspended above the boiling liquid. Typically, the most accurate boiling point measurement...

...WORD ASSOCIATION TEST Experiment No. 2 NAME: CRISELLE M. MACASPAC SCORE: DAY/TIME: W 7:30-4:30 DATE: TITLE: Word Association Test ABSTRACT: The formation of word association and perceptual defense aim to measure signs of emotional complexes and to be able to identify the degree of threshold for each selected stimulus. The apparatus needed were paper, pencil and ninety-nine (99) stimulus words. For the procedure, the experiment has 1 phase: determination of association reaction time and the measurement of recognition thresholds. Ninety-nine carefully chosen words were presented one at a time. The experiment concludes that there were different signs of emotional complexes and different degree of threshold for each selected stimulus words. Because the subject had difficulty in reacting to certain stimulus word, the subject felt unpleasant with the stimulus words so the individual laugh. INTRODUCTION: Perceptual defense not only do absolute threshold vary from person to person; they also vary from time for a single person. The type of stimulus, the state of one’s nervous system, and the cost of false “detections” all make a difference. Emotional factors are also important. Unpleasant stimuli, for example, may raise the threshold for recognition. This effect is called perceptual defense. “Dirty” words took longer to recognize when flashed on a screen that did “clean” words. Apparently, it is possible to process information on more than one level and...
