Task 1 A. Compare your observations from the four activities of the fresh wet mount, direct staining using crystal violet, and indirect staining using both Congo Red and crystal violet.
When the wet preparation of slides containing Saccharomyces cerevisiae (yeast cells) was observed under x10 and x 40 magnification, yeast cells appeared as independent oval shaped structures. In the wet mount, all the cells had the same appearance and their borders were not clearly distinct. It was very difficult to determine the shapes of epithelial cells in the wet mount. In direct staining, the shapes of the cells were clearly visible. In direct staining, the background of the cells appeared violet in color making it easy to be able to identify the shape of cells. Indirect staining with Congo red dye made the cells appear reddish in color. Congo red dye stained the cells leaving the background without dye. The shapes of S. Ceverisae were distinct, but the epithelial cells appeared reddish with no clear distinction. Crystal violet staining produced the best results because it dyed the background making the outline of the cells clearly distinctive (Estridge, Reynolds, & Walters, 2007). B. Discuss whether you were able to identify specific bacterial morphologies. In crystal violet staining coccal (circular) shaped bacteria were observed. These bacteria were independent (not filamentous). In the wet mount, bacterial morphology was difficult to identify (Estridge, Reynolds, & Walters, 2007). C. Explain the difference between direct and indirect staining. Direct staining was done with basic dyes like crystal violet which colors the cell and it contains positive ions that attracts negative ions of the bacteria cells, while indirect staining was done using acidic dyes like Congo red dye.
