All three lac operators were mutated by site directed mutagenesis and were infected into strain CHS 9 rec A (i^+,z^-) cells. Results of β-galactosidase assay under repressed and induced conditions showed an almost total loss of repression when O1 is destroyed. When either O2 or O3 is inactivated there was slight decrease of repression, but a 70-fold decrease of repression was seen when both pseudo-operators combined were lost.
