A clean, dry, Perspex gel casting moult was sealed with adhesive tape on both sides and kept it on a leveling platform, after putting a comb on the slot. 2. 1X TAE Electrophoresis buffer was aliquoted 100ml into a conical flask and 1gm of agarose was added to it, and was boiled untill the agarose melts completely in a microwave oven. 3. Solution was cooled to 60°C, and ethidium bromide was added (stock solution of 10mg/ml) to a final concentration of 0.5mg/ml and mixed thoroughly. 4. Warm agarose solution was poured into the mold. The thickness of gel should be between 3mm to 5mm.There should be no air bubbles under or between the teeth of the comb. 5. After the gel was set completely the comb and the tape were carefully removed and the gel was mounted in the electrophoresis tank, filled with electrophoresis buffer (1X TBE). 6.
