...MLT1 – Lab Report 1 Bacterial Morphology Nichole Performed at Home 3/21/13 Bacterial Morphology A. Comparing Observations from Mounting Techniques Four mounting techniques were used during this experiment which included the fresh wet mount, direct staining using crystal violet, and indirect staining using both Congo Red and crystal violet. There was a liquid appearance in the wet mount slides with the cells appearing very similar in color to the liquid from the sample. The cells and their borders in the wet mounts were not as distinct in appearance as in the staining methods. While the bacteria shapes were able to be identified in the yeast wet mount, it was very difficult to determine the shapes in the cheek smear. The next method used was the direct staining using crystal violet. The cells in the direct stained slides were very easy to view, well defined and were a distinct violet color. The indirect staining method using the Congo Red dye appeared to actually stain the cells in the cheek smear rather than the background and area around the cell. In the higher resolution cheek cell slide, the cell was outlined with the Congo Red dye. The yeast cells were easily identified using the indirect method however the cells in the plaque smear were not. There was a large area of dye in one area on the slide...

According to previous study, approximately 75% women have vulvovaginal candidiasis at least once in their live, while 40-50% of them develop 2 or more than two episodes (1). Another common cause of abnormal vaginal discharge, bacterial vaginosis (BV), also have a considerably high prevalence (1). Since Rebecca was having unprotected vaginal sex with her male partner for 9-months, an un-safe sex preference must be considered in this situation. Which means, Rebecca is likely to have unprotected sex with her casual male partners (e.g. no condoms), and STIs must be considered. Trichomonas vaginalis is a common bacterial cause of abnormal vaginal discharge (1), also the commonest non-viral STI across the world (2), it should be considered in this case. This was addressed by observing the changes in cells as they were surrounded by NaCl solution and later by a water solution. The results support our hypothesis and show that the composition of the surroundings has a visible (under a microscope) effect on the shape and structure of each individual cell. As the NaCl concentration increased, water rushed out of the cells and caused them to pull away from the wall while as the water concentration increased water rushed into the cells and caused them to swell. Introduction: The “big problem” that is being addressed is the change in a cell’s structure when surrounded by different concentrations of solutions. It is known that substances move from areas of high concentration to areas of low concentration, but to validate this knowledge, we must observe the change in a cell from its “normal” state with the addition of solutions. Questions that must be answered are what affect a different NaCl solution has on plant cells as compared to water? The hypothesis being tested in this report is that when a plant cell is introduced to a NaCl solution, the water in the cells will move out to an area of low concentration, thereby altering the shape of the cell. ...HISTORY OF BAKING On today’s market there is a never ending variety of the most delicious sweets and pastries to please both the palate and the eyes. We have become so used to this diverse range of bread, mouth watering pastries and cakes that seldom we ask just how they come into existence. I guess my initial interest in the origin of the baking industry was not aroused by accident; rather I suspect it was kindled by a chain of events occurring earlier in my life... My Grandfather owned a small hotel and bakery in Switzerland and when he retired my father who was an excellent pastry cook continued the business. As a small child I spent many hours watching my dad at work and later followed in his footsteps almost as a matter of course. Then again, I have a suspicion that it may have been “fate-by-design” that made me take up pastry-cooking. The question still remains, was my career pre-determined by the fact that my grandfather had thousands of letterheads printed for his bakery which included his name? The prospect of a hefty quantity discount would no doubt have made perfect sense to my frugal fore bearer; it must have been so much cheaper getting all this printing done in a big way. Grandfather should have realised (and I suspect that he might have) that there was not the slightest chance that any one generation could ever use up this huge mountain of stationery. However, in the past 40 years, the number of cases of cervical cancer and the number of deaths from cervical cancer have declined radically. This decrease is largely the result of many women getting regular Pap tests (Center for Disease Control and Prevention, 2004). Persistent infection with high-risk human papillomavirus (HPV) infections can lead to cervical cancer. Since HPV and precancerous lesions of the cervix are usually asymptomatic, prevention and regular screening remains imperative for early detection and treatment of cervical cancer. Here we examine strategies for prevention, assessment, and management for cervical cancer and contemplate briefly potential implications if left undiagnosed or untreated.
