Enzymes are catalytic proteins, that when present in a chemical reaction, are able to lower the action potential needed to create the reaction without being destroyed or altered themselves in the process. In Part A, my hypothesis stated that when Catalase is combined with H2O2 the rate of conversion to water and oxygen gas should double when 5-10 drops of enzyme is added and quadruple when 10-20 drops are added. In Part B, my hypothesis stated that increases in enzyme concentration or buffer pH the substrate of the final product will yield increased substrate, also, if the substrate concentration is increased then the enzyme will be less diluted, the buffer pH will increase, or there will be a temperature increase. In Part C, my hypothesis stated that tacrine will have an inhibitory effect on AChE, and that those effects will increase as the level of concentration increases. In all three labs I postulated that increases in temperature and concentration levels and would increase the rates and decrease time to form chemical reactions. The heart rate (HR) is each full beat, or each pump, the heart does; this is recorded in beats per minute (bpm). The blood pressure (BP) is the amount of resistance the heart works against the arterial walls during each pump. My hypothesis for this lab was that my subject would have the same HR and BP for each of the locations and different methods used. I stated this because, at rest, the heart should be working at the same level no matter the position; although one position might slight lessen the heart’s workload. This is important in terms of my subject’s health and fitness because it means he has a normal, young strong heart. Per minute, it is important for the numbers to be lower because it means his heart is not working too hard but can supply his body with a sufficient amount of blood. Determine Taster Genotype Hypothesis: If I am a taster, then my genotype for PTC taster must be either TT (homozygous dominant) or Tt (heterozygous) I – Results: This experiment aimed to investigate the allele frequency of the PTC taster gene (TAS2R38) in a small population, represented by the students in class. The genotype obtained from genomic analysis (via PCR and gel electrophoresis) confirmed that the genotypic result is consistent with the phenotypic result observed at the beginning of the lab. However, DNA fragments of 3 lab subjects didn’t show up on the gel. The allele frequencies can’t be calculated because the data is insufficient to apply the Hardy-Weinberg equation. There are many factors that might be contributed to the invisibility of these DNA fragments, most likely accidental errors. For example, the DNA wasn’t loaded onto the gel probably, or the DNA for some reason didn’t sink to the bottom of the well, or just simply there was not enough DNA. To determine the genotypic profile of the students PTC gene, DNA samplers from each individual was collected from saliva. Using premade PTC primers (short oligonucleotides), a DNA template that encoded the PTC gene (approximately, 303 bp) was amplified by PCR.
