The positively best bacteria to receive when doing an unknown project

The word bacteria receives a bad connotation in the everyday world but some may not realize the importance bacteria has on a person’s immunity and health. There can be both good and bad bacteria. Good bacteria, for example, helps to breakdown food enabling the digestion process to work smoothly and absorb nutrients like probiotics. These good bacteria are often found in foods like yogurt, cheese, and sauerkraut. Best of all, good bacteria can help treat infectious diseases. A study has shown that, when injected with a good predatory bacteria like Micavibrio aeruginosavorus and Bdellovibrio baceriovorus, an antibiotic resistant bacteria was defenseless (Nordqvist, 2013). The study was created for an eye infection to see whether good bacterial pathogens would be able to fight off bad bacterial pathogens without damaging the eye or causing further irritation. The term microbiology means the study of microscopic organisms. Within the specialized area, microbiologists help to identify new organisms and how they affect life on earth. There are new organisms being discovered every day, and there could be dangers or losses without proper identification of how they live, what they do, and can they be controlled. A study was done to show the process of categorizing and identifying an unknown organism. Throughout the semester, multiple tests were performed to distinguish between the different types of bacteria and characteristics that make them standout from similar bacteria. Since microbes are the longest living things on earth, the study of microbes should be important and become a priority for many since these small organism do not have to be seen in order to kill. These microscopic pathogens can serve for the better or worse.
Materials and Methods
The first day of lab began by randomly receiving an unknown organism labeled A6. In order to identify its gram reaction, a gram stain was performed (Leboffe & Pierce, 2011). Once the gram stain was finished, the unknown was observed using a microscope at a 1000X magnification and the results were recorded. By using aseptic techniques, the following inoculations occurred: a nutrient broth while using a loop, a nutrient agar slat while using a needle, and a nutrient agar plate using the four quadrant streak method with a loop (Leboffe & Pierce, 2011). When inoculations were completed and properly labeled, they were placed in the section reserved for organisms to be incubated at 37°C. To confirm the results of the gram stain, a KOH test was also performed using a 3% potassium hydroxide (Leboffe & Pierce, 2011). The morphology and gram was confirmed by the instructor and Table 1 provides tests, purpose, reagents and results.
The second day of lab began by recording the results of the broth, plate, and slant from the previous day of class. After the results were recorded, using aseptic techniques, the following test media was inoculated using the unknown organism grown on the nutrient agar plate: blood agar plate, mannitol salt agar plate, phenol red mannitol broth tube, nutrient gelatin tube, MRVP broth tube, phenol red glucose broth tube, urea agar plate (Leboffe & Pierce, 2011). Each inoculated media was properly labeled and was placed in the section reserved for organisms to be incubated at 37°C except for the nutrient gelatin tube as it was incubated for seven days at room temperature. Table 1 provides tests, purpose, reagents and results.
The third day of lab began by starting the MRVP test first; this was done due to time constraints (Leboffe & Pierce, 2011). While observing the MRVP test, records were taken of the results from the tubes and plates incubated the previous day of class and then properly discarded. Then, using aseptic techniques, the following test media was inoculated using the unknown organism grown on the nutrient agar plate: tributyrin agar plate, citrate slant, motility stab, nitrate broth tube, and SIM tube (sulfur reduction, indole production, and motility) (Leboffe & Pierce, 2011). Each inoculated media was properly labeled and was placed in the section reserved for organisms to be incubated at 37°C. Table 1 provides tests, purpose, reagents and results.
The fourth day of lab began with completing the nitrate reduction test by adding eight drops reagent A and reagent B to the tube and recorded the results. Also, the SIM test was completed by recording the results and observations, then by adding ten drops of Kovac’s reagent to the tube, and finally recording the results after a wait period of three minutes. After completing the SIM test, a catalase test was performed by using the unknown cultures taken from the first day and aseptically placed a large loopful of unknown bacteria on a clean microscope slide. Then three to ten drops of hydrogen peroxide were added to the bacteria covered slide; results were recorded and slide was properly discarded. Once the catalase test was complete, an oxidase test was performed by using a Dry Slide and transferring a large amount of unknown bacteria using a sterile swab on to one square of the Dry Slide. After twenty seconds, results and observations were recorded and Dry Slide was properly discarded. Records were then taken of the results from the tubes and plates incubated the previous day of class and then properly discarded; that included the gelatin tube as the test had to be incubated for seven day. Lastly, using aseptic techniques, the following test media was inoculated using the unknown organism grown on the nutrient agar plate: Milk agar plate and starch agar plate (Leboffe & Pierce, 2011). Each inoculated media was properly labeled and was placed in the section reserved for organisms to be incubated at 37°C. Table 1 provides tests, purpose, reagents and results.
The fifth day of lab began by obtaining the starch agar plate from the previous day of class and adding enough Gram’s iodine to flood the plate. After two minutes, results and observations were recorded and plate was properly discarded. The results of the milk agar plate from the previous day of class was observed and recorded then properly discarded. Table 1 provides tests, purpose, reagents and results.
Results
After the inoculation of unknown A6 on the nutrient agar plate, the morphology was observed as small, round, circular colonies with a smooth white/yellowish color. The inoculation of the unknown on the slant agar showed it to be filiform while the inoculated broth showed turbidity with sediment. After the gram stain test confirmed the unknown to be a gram positive, cocci bacteria. Table 1 lists all of the test performed, what each test would provide, any reagents used, and the results received after performing each test on the unknown bacteria A6. Multiple tests were performed but the flow chart categorizes key tests which assisted with the identification of unknown A6.

Discussion
As specified on the flow chart, the test performed concluded that the unknown was identified as Staphylococcus aureus. After determining the bacteria to be gram positive as well as a cocci instead of a bacilli, the list of unknowns were reduced by 75% leaving me with six possible unknowns. There were two tests which helped narrow down the list further: Blood Agar and Catalase. During the catalase test, the bacteria created bubbles after the addition of hydrogen peroxide which identified that my unknown had catalase, an enzyme that breaks down hydrogen peroxide into water and oxygen. This reduced the list of six to only three. After the Blood Agar test, the bacteria resulted to a complete hemolysis of red blood cells which was represented by clearing around the grown colonies. The known test results of two out of three unknowns remaining have no hemolysis of red blood cells which left S. aureus as the last bacteria.
S. aureus comes from the genus Staphylococcus and are small, circular species which tend to form small clusters. This bacteria was first discovered in pustules from a surgeon in Scotland around 1880 by Sir Alexander Ogston (Staphylococcus, 2014). One form of this infection is found on the skin as pimple like abscesses which can grow excessively and require surgical draining if not cared for quickly and properly. S. aureus is more widely known from the infection MRSA, Methicillin-resistant Staphylococcus aureus, which is a bacterial strain of Staphylococcal that becomes resistant to antibiotics which is found mostly in medical care type facilities (Chambers, 2001).
