The scientific study of microorganism is known as microbiology. Microorganisms are small living things or organisms that cannot be seen without the use of a microscope. The microorganism can be categorized as eukaryotes for instance plant, animal, protozoa, fungi and they can also be grouped into prokaryotes such as bacteria and archaea. This paper will focus on how to identify an unknown bacterium. Bacteria which is also known as the bacterium is a non-cellular microbe that lacks membrane organelles, nucleus, and cell walls. Although bacteria are very small, they have a large diversity and they are different in shapes and sizes. Bacteria have three shapes which are bacilli (rod) shaped, cocci (round) shaped and spirilla (spiral) shaped. The …show more content…
This test helps identify the color and the shape of the bacteria under microscopic lens which aids in determining whether the bacteria is gram negative(pink) or gram-positive (purple) (Silby, 2017). The second test that was performed in this lab is the oxidase test which helps determine the presence of cytochrome and it distinguishes between bacteria that makes use of terminal oxidase and the once that uses cytochrome oxidase (Silby, 2017). The third test that was performed in this lab is fermentation test which is used to determine whether the isolate can ferment the three sugar which are dextrose, mannitol due to the fact that bacteria often manufacture acid when fermenting sugar and knowing the pH is the best way to see if they ferment. It also tests to see if an organism can use a certain carbohydrate and it also test for the presence of gas and acid yield from fermentation. The third test that was performed is Indole test which the ability of organisms to be able to convert from tryptophan to indole (Silby, 2017). The fourth test that was done in this lab is the methyl red and Voges Proskauer (MRVP). The methyl red test was done in other to determine if bacteria have the ability to acid from glucose metabolism. If the test shows a positive, then the bacteria uses oxygen as electron acceptor. I got a wood stick and rubbed some of my bacteria onto the oxidase card. Within 20 seconds the oxidase card turned blue indicating a positive result. This also explains my result from the gas pack test. Oxygen is important to my bacteria. All known organisms can be lined up by their size. To figure out what size the bacteria I did a simple stain of my bacteria to figure out the size of it. Inspecting the slide under the microscope, it was revealed that my bacteria is less than 2um. Viruses are the smallest microorganism out there ranging from 20nm to 300nm. They are invisible to the naked eye. An electron microscope is needed to visualize them. Acid and base test helps to know which type of environment is beneficial to a bacteria. To figure out my family I had to inoculate a 3.6pH broth to see if my bacteria can survive acidic environment. I inoculated the broth and placed it into a 37C incubator for 48 hours. When I came back to inspect the broth, I found there was no growth. This test finally helped me to narrow down the bacteria family, which is Pseudomonas.
